The BD Rhapsody Single-Cell Analysis System is a microwell-based single-cell partitioning system that allows users to retrieve multi-omics data of up to 40’000 cells per cartridge, including expression of up to 200 surface proteins, mRNA, and VDJ sequences of T cell and B cell receptors. The expression of surface proteins is detected through antibodies conjugated to oligonucleotide tags. It is possible to multiplex samples, loading cells from up to 12 or 24 samples into one cartridge.
In addition, the BD Rhapsody Scanner provides visual quality control metrics including number of cells loaded, viability and multiplet rate, enabling users to take an informed decision about whether to proceed or not with cDNA synthesis and (expensive) downstream sample processing.
Using the BD Rhapsody Express Single-Cell Analysis System, users can load cells into cartridges and proceed up to cDNA synthesis and exonuclease treatment in the Cytometry Facility. Afterwards, the resulting beads with cDNA can be stored at 4°C for later library preparation and sample sequencing in other laboratories. For users sending samples to the Functional Genomics Center Zurich (FGCZ), a meeting with staff from the Cytometry Facility and the FGCZ can be arranged to coordinate the downstream processing of the samples following cDNA synthesis.
The system is located at Y44G03. The BD Rhapsody Express Single-Cell Analysis System can be easily transported to other laboratories to shorten the time between sample preparation and cell loading into the cartridge (the system is installed on a mobile desk which simplifies transport within the Irchel campus).
Biosafety note: the system runs on a biosafety 2 permit. Please treat the workspace accordingly and indicate the biosafety level of your experiment in the instrument booking system.
